Two people having different idea’s/opinions about something can be the start to something amazing. Its the way you debate your ideas and accept being wrong in a proper way that takes the talk to the right or wrong path. Its ok to be wrong if you accept it and have learned something. The minute name calling enters a debate the whole learning something goes out the door in many ways. I think if I proved someone with a phD wrong I would silently take the victory, shake hands and look forward to the next debate so I can learn or teach more. I couldnt imagine if a professor had a student that had a wrong answer or opinion and the teacher started calling that student names. That is the wrong way to teach/learn. Ego is your enemy. I am probably one of those “half the guys around here” and proud to be on that half if someone with such an abrasive way of teaching is on the other half.

My understanding is that calculated logP is a useful tool for designing compounds that you’re considering synthesizing but is also considered pretty unreliable. Different logP programs come up with different values. I don’t think showing the calculated logP really is the debate ender that it is presented as.

You could just do the experiment, I think. It’s the partition coefficient between water and octanol. That’d really put the cards on the table so to speak. Surely KoK has samples of d9 and THCO around to do it.

I would do this but idk how to calculate log p, I read somewhere that computational log p is usually pretty accurate, the computer could be wrong but based on what people have been telling me and what I’ve read thco has a lower log p. The longer duration proves this IMO. I’d love to do an actual experiment and test plasma levels, acetylation of cannabinoids is so new theres nothing available on bio availability really. One patent does claim longer duration which my clients are seeing.

One thing I’m working on is a cannabinoid for heroin addiction and I think cbd diacetate is the key. One of my good friends sisters is super stuck on heroin and cbd surprisingly is the only thing that helps her. The only thing is it doesn’t last very long.

According to that program diacetylating cbd would lower its log p from 7.75 to 5.9 which would be HUGE for bioavailability. Idk acetylation isn’t necessarily going to make a perfect pro drug, it does increase duration though which helps me sleep threw the night when I consume thc o instead of thc.

My cousin is in his 4th year at uc Davis and he’s specializing in molecular biology for cancer drug development. I’m going to spend some time talking with him about all these different subjects.

One of the silver lining outcomes of this was that I had to revisit meta studies on bioavailability modeling. And I found an instance in which THC was in the dataset. That dataset listed THC’s calculated logP as being 5.53 meaning that partitioning between n-octanol and water is predicted to result in a concentration in the octanol phase being 10^5.53 ~ 340,000 higher than that of the concentration in the aqueous phase.

You’re absolutely right about the range of values obtained by different algorithms but it’s better than nothing. To experimentally and accurately determine a half a million to one ratio is hard and fraught with is own errors.
