How much spore water do I add to each bag (2 pounds) and mason jar (pint)? Also do I mix the bag or do I wait?

3-5mL should be enough. Roll the bag around the first few days to make sure theres no pooling liquid.
If you want to guarantee success, try innoculating some petri dishes instead of the grain bag.
Spore to grain is risky because spore syringes usually arent sterile like liquid culture syringes
Completely agree, or at the very least be prepared to clone a viable piece of fruit since the bags you get will be what is called a multispore culture and an isolation is wanted
You can recover any culture within 3 platings over 14 days.

Plates are a visual checksum: plate until it’s ropey. No greens, fuzz, or anything other than rhizo.
Then you can throw a slice and continue knowing you didn’t drag along unwanted cultures.
Edit: remember plates on agar do best when not allowed to grow to the edge. Best to make selections sooner rather than later. You can slow or stall growth in a wine fridge or freezer.

I’m going to guess malt extract agar, potato dextrose agar, not quite sure of the other two
Y is yeast. I’ve seen some great growth with a dash of nutritional yeast in my agar.
